Противоопухолевое действие индукторов ферроптоза на органоиды метастатического колоректального рака

Аннотация
На сегодняшний день колоректальный рак (КРР) остается одним из наиболее распространенных онкологических заболеваний с высокими показателями смертности во всем мире. Прогресс последних десятилетий в изучении механизмов, лежащих в основе лекарственной устойчивости КРР, указывает на необходимость поиска новых терапевтических подходов. Так, известно, что опухолевые клетки, не восприимчивые к стандартной терапии и к известным механизмам запрограммированной клеточной гибели, таким как апоптоз, чувствительны к ферроптозу. В связи с этим индукция ферроптоза рассматривается в качестве альтернативной стратегии для разработки противоопухолевых лекарственных средств. На культурах органоидов метастатического КРР, ранее полученных из опухолей трех пациентов, нами изучено действие известных индукторов ферроптоза: эрастина и сорафениба, ‒ как в виде монотерапии, так и в комбинации со стандартными химиотерапевтическими препаратами для лечения КРР. Показано, что эти индукторы ферроптоза обладают противоопухолевой активностью. Кроме того, в ряде случаев эрастин и сорафениб усиливали действие стандартных химиотерапевтических препаратов на культуры органоидов метастатического КРР. Таким образом, дополнительная к стандартной терапии индукция ферроптоза в опухолевой ткани КРР может быть перспективной терапевтической опцией и заслуживает дальнейшего изучения в доклинических и клинических условиях.
