Адаптация ацидофильного гриба Sistotrema brinkmannii к рН фактору

Аннотация
Исследование скорости роста гриба Sistotrema brinkmannii при различных значениях рН среды, температуры и концентрации NaCl показало, что он является мезофилом, предпочитает бессолевую среду и относится к облигатным ацидофилам, так как имеет выраженный оптимум роста при рН 3.0–4.0 и не растет при рН 7.0. Для изучения защитных механизмов, позволяющих грибу развиваться в кислых условиях среды, был исследован состав его осмолитов и липидов. Впервые показано, что в мицелии гриба в динамике роста в оптимальных условиях присутствует большое количество трегалозы (4.0–6.6% от сухой массы), что доказывает участие осмолитов в адаптации у ацидофилов. При этом на границах диапазона роста (рН 2.6 и 6.0) количество трегалозы в мицелии гриба снижалось в 2.5 раза, что соответствует узкому оптимуму роста гриба в природных условиях (рН 3.0–4.0). Отличительной чертой состава мембранных липидов гриба является высокая доля сфинголипидов (до 60% от суммы), которая в динамике роста в оптимальных условиях снижается вдвое. К основным мембранным липидам, кроме сфинголипидов, также относятся фосфатидные кислоты, фосфатидилэтаноламины и стерины, доля которых с возрастом увеличивается. Состав мембранных липидов гриба при рН 2.6 практически не отличается от оптимальных условий, тогда как в околонейтральной области наблюдается двукратное повышение доли сфинголипидов, что указывает на их адаптивное значение. Одновременное снижение доли сфинголипидов и рост уровня трегалозы в динамике роста позволяет предположить взаимосвязь этих соединений в защите мембран клетки.
