ПОИСК ВАКЦИН И ТЕРАПЕВТИЧЕСКИХ НУКЛЕИНОВЫХ КИСЛОТ ПРОТИВ РАКА

Аннотация
В статье кратко рассматриваются особенности иммунной системы при канцерогенезе и причины утраты ею надзора над раком. Для поиска в первичной структуре белков, ассоциированных с раком, нонапептидов (иммунных эпитопов), распознаваемых главным комплексом гистосовместимости типа I, предложен биоинформационный подход. Он принципиально отличается от других биоинформативных инструментов определения иммунных эпитопов тем, что дифференциация пептидов построена на учете их первичной структуры, что невозможно учесть при использовании методов, основанных на использовании физико-химических характеристик аминокислот. В качестве полезного этапа дизайна вакцины может быть компьютерное моделирование процессинга белков в иммунопротеосомах, позволяющее предвидеть вероятность сохранности иммунного эпитопа для презентации его Т-клеткам. Анализируются проблемы в поисках вакцин и критически оценивается возможность создания универсальной вакцины против разных типов рака на основе общих для них антигенов, так как невозможно представить в одной вакцине многообразие мозаик мутационных изменений каждого пациента и синтезировать, например, мРНК, как наиболее приемлемую форму вакцины для дизайна универсальной вакцины, без нарушения ограничений в ее синтаксисе. Оценены перспективы и проблемы использования терапевтических нуклеиновых кислот (малые интерферирующие РНК, микроРНК, антисмысловые олигонуклеотиды, мРНК, аптомеры CRISPR-Cas9 система редактирования гена) против рака, расширяющих возможности химиотерапии и вакцинологии.
