Геномная характеристика бактерий Bacillus subtilis PY79 и NCIB 3610 как потенциальных продуцентов сурфактина

Аннотация
Сурфактин является одним из мощных поверхностно-активных веществ биологического происхождения и обладает широким спектром потенциальных сфер применения в промышленности и медицине. Главным фактором, ограничивающим его активное использование, является высокая стоимость производства. Из-за сложности структуры химический синтез сурфактина нерентабелен, поэтому основным подходом к его получению является микробиологический синтез. В данной работе способность синтезировать сурфактин продемонстрирована для штамма Bacillus subtilis NCIB 3610, обладающего свойствами, потерянными большинством модельных лабораторных штаммов B. subtilis. Методом нанопорового секвенирования получены последовательности геномов B. subtilis NCIB 3610 и B. subtilis PY79 и проведен их сравнительный анализ с геномом штамма B. subtilis 168, генно-модифицированные производные которого являются запатентованными продуцентами сурфактина. Показано отсутствие в штамме NCIB 3610 каких-либо мутаций, способных помешать биосинтезу сурфактина. В штамме PY79 выявлены мутации в генах, участвующих в клеточных процессах, конкурирующих с биосинтезом сурфактина. Таким образом, полученные в данной работе результаты позволяют рассматривать B. subtilis NCIB 3610 и PY79 как альтернативные штаммы-основы для генно-инженерного конструирования потенциального продуцента сурфактина.
