Транскриптомный анализ штамма Levilactobacillus brevis 47f в условиях окислительного стресса

Аннотация
Levilactobacillus brevis 47f – гетероферментативная аэротолерантная молочнокислая бактерия, выделенная из микробиоты желудочно-кишечного тракта здорового человека. В комплексе доклинических исследований штамм проявил противовоспалительные свойства в отношении мукозита слизистой оболочки тонкого и толстого кишечника, индуцированного 5-фторурацилом. Вместе с тем молекулярные механизмы, определяющие свойства штамма и, в частности, его реакцию на воздействие активных форм кислорода, остаются неизученными. Задачей данной работы было исследовать реакцию штамма на действие окислителей – перекиси водорода и кислорода – методом транскрипционного RNAseq-анализа. Оба окислителя оказали сильное действие на штамм, увеличивая или снижая экспрессию нескольких сот генов – как общих, так и специфических для каждого оксиданта. Приведена характеристика белков с наиболее увеличенной экспрессией (ДЭ ≥ 5). Гены, активизирующиеся при действии обоих оксидантов, кодируют белки стресса, антиоксидантной защиты, репарации белков и нуклеотидов, клеточной стенки, транспорта и метаболизма углеводов, катаболических путей запасания энергии. При действии перекиси активизируется транскрипция в основном белков защиты: стрессового ответа и молекулярных шаперонов, антиоксидантной активности, репарации ДНК, а также белков, участвующих в формировании клеточной стенки. В аэробных условиях в значительной степени активизируются гены, определяющие белки преобразования энергии (использование в качестве источника энергии, помимо глюкозы, фруктозы, жирных кислот, нуклеозидов; белки фосфокетолазного пути) и импорта пептидов, аминокислот, сахаров. Полученные в настоящей работе результаты будут использованы нами при интегрированном анализе совокупности транскриптомных, протеомных и метаболомных данных по данному штамму. Это внесет значительный вклад в создание на основе L. brevis 47f фармабиотика для лечения различных воспалительных заболеваний.
